Sterile syringe filters, including gas filters or liquid filters, should be tested for integrity according to the sterile filter integrity test procedures before and after use. After the integrity test, the membranes and filters used for aseptic production need to be sterilized. The sterilization methods of sterilized syringe filters are divided into steam sterilization and ethylene oxide gas sterilization. The general method is steam sterilization. For in-line steam sterilization of sterile syringe filters, the differential pressure before and after the filter should be monitored during sterilization.

The general sterile syringe filter should be kept in an upright position, and should not be placed obliquely or lying down. The sterilization time, pressure and temperature should be recorded during the sterilization process. A sterile syringe filter cannot filter different products at the same time, but can be used to filter multiple batches of the same product, but must be tested for integrity. A sterile syringe filter can be used for no more than 1 working day. In the process of use, if the filtration rate suddenly becomes faster or too slow, it means that the membrane has been damaged. Then replace it with a new membrane and do the experiment again.

Precautions for the use of sterilized syringe filters

1. Do not use a syringe smaller than 10cc, because the small volume of the column tube may cause the pressure to exceed the upper limit, resulting in damage to the filter membrane or personal injury.
2. In the laboratory, it’s for one-time use.
3. Part of the filtrate at the beginning should be discarded. Taking 25mm as an example, the first 1mL of the filtrate should be discarded or the filter should be pre-cleaned with 1 to 2mL of solution.
